This is the most popular way to say “lock the door.”
In a daily conversation, we often omit “を.”
We say “鍵(かぎ)掛(か)ける.”

鍵(かぎ)をしめる
鍵(かぎ) = key
しめる = close
We say “鍵(かぎ)をしめる (close the key)” which means “lock the door.”
